IL-4 and IL-17 Are Required for House Dust Mite-Driven Airway Hyperresponsiveness in Autoimmune Diabetes-Prone Non-Obese Diabetic Mice

چکیده

Allergic asthma is characterized by airway inflammation with a Th2-type cytokine profile, hyper-IgE production, mucus hypersecretion, and hyperreactivity (AHR). It increasingly recognized that heterogeneous disease implicating complex immune mechanisms resulting in distinct endotypes observed patients. In this study, we showed non-obese diabetic (NOD) mice, which spontaneously develop autoimmune diabetes, undergo more severe allergic AHR than pro-Th2 BALB/c mice upon house dust mite (HDM) sensitization challenge. The use of IL-4-deficient NOD the vivo neutralization IL-17 demonstrated both IL-4 are responsible exacerbated mice. Overall, our findings indicate diabetes-prone might become useful as new HDM-induced model to elucidate dysimmune involving Th2 Th17 responses could better mimic some asthmatic endoytpes.

Vaccination against IL-33 Inhibits Airway Hyperresponsiveness and Inflammation in a House Dust Mite Model of Asthma

In several clinical and experimental studies IL-33 and its receptor have been found to play important roles in the development of asthma and allergic airway inflammation. We evaluated the effects of vaccination against IL-33 in a mouse model of airway inflammation induced by house dust mite (HDM) allergen. Balb/c mice received the IL-33 vaccine subcutaneously, followed by intranasal administrat...

Roles of Bronchopulmonary C-fibers in airway Hyperresponsiveness and airway remodeling induced by house dust mite

BACKGROUND Asthma is characterized by chronic airway inflammation, airway hyperresponsiveness (AHR), and airway remodeling. While exposure of house dust mites (HDM) is a common cause of asthma, the pathogenesis of the HDM-induced asthma is not fully understood. Bronchopulmonary C-fibers (PCFs) contribute to the neurogenic inflammation, viral infection induced-persistent AHR, and ovalbumin induc...

appears on the skin. We separated lymphocytes from the inflamed skin of NC/ Nga mice using collagenase I (Sigma-Aldrich) and counted the numbers of nuocytes (ST21/Sca-11/lineage marker-negative) by FACS. Results: (1) The reactions were hapten specific. Wild-type BALB/c mice showed both immediate and delayed-type reactions, whereas ST2 KO mice did not show any immediate reaction. (2) When IL-33 ...

Selective stimulation of IL-4 receptor on smooth muscle induces airway hyperresponsiveness in mice

Production of the cytokines IL-4 and IL-13 is increased in both human asthma and mouse asthma models, and Stat6 activation by the common IL-4/IL-13R drives most mouse model pathophysiology, including airway hyperresponsiveness (AHR). However, the precise cellular mechanisms through which IL-4Rα induces AHR remain unclear. Overzealous bronchial smooth muscle constriction is thought to underlie A...
